摘要
目的探讨链脲佐菌素(STZ)建成可靠、稳定的1型大鼠糖尿病模型情况。方法选取SDF级雄性Wistar大鼠70只,分成3个实验组,各20只和对照组10只,实验组大鼠一次性腹腔注射STZ 30、65、100 mg/kg,对照组10只大鼠腹腔注射枸橼酸缓冲液,用拜安易血糖仪检测血糖,7 d后血糖>16.65 mmol/L判定为1型糖尿病动物模型。观察大鼠的血糖、体质量、饮水量、胰岛素和C肽等指标。结果大鼠注射STZ 65 mg/kg 7 d后,大部分血糖值达到成模标准,并出现糖尿病表现,持续观察7周,有18只大鼠成模,未见糖尿病转复,糖尿病症状明显。结论腹腔一次性注射STZ65 mg/kg剂量可成功制备1型糖尿病大鼠模型。
Objective To establish type I diabetic rat models by intraperitoneal injection of streptozoto- cin and observe the stability of diabetic changes in rats. Methods 70 male Wistar rats were divided into 3 experiment groups of 20 rats each and 1 control group of 10 rats. The experiment groups were intraperitoneally injected by 30 mg/kg,65 mg/kg, and 100 mg/kg STZ respectively, while the control group was injected cit- rate buffer. Bayer blood glucose meter was adopted to test the blood glucose, setting 〉 16.65mmol/L after 7 d as the type 1 diabetic animal model. The fasting blood glucose level, body weight, water intake, insulin and C-peptide of the rats at different time points were observed/measured. Results In the group of injection by 65 mg/kg STZ, after 7 days mast rats reached the model blood glucose standard and had diabetic manifes- tation, after continuous observation of 7 weeks, 18 rats were qualified as the model with obvious symptoms and without conversion. Conclusion With the dose of 65 mg/kg of STZ through one intraperitoneal injection, type 1 diabetic rat models can successfully established.
