摘要
比较研究了在 PEG水相和 PEG甲苯有机相中辣根过氧化物酶 (HRP)的 U V- Vis光谱 .研究表明 ,PEG甲苯体系破坏了酶的结构并导致血红素辅基脱落 .利用停流光谱仪对 HRP与过氧化氢复合物的形成进行研究 .结果发现 ,PEG甲苯体系中复合物 的形成比 PEG水溶液中复合物 的形成慢 4倍左右 ,并且在 PEG甲苯体系中 ,复合物 很不稳定 ,导致了酶失活 .
The UV Vis spectra of horseradish peroxidase (HRP)and its compound with hydrogen peroxide(HRP Ⅱ) in PEG toluene solution were studied by comparision with their UV Vis spectra in PEG water solution. It was suggested that PEG toluene disrupted the enzyme structure and caused the heme group to depart the enzyme. In both reaction media, the HRP compounds with hydrogen peroxide were also investigated. It was found that the formation of HRPⅡ is about 4 times faster in PEG water solution than that in PEG toluene solution, and HRPⅡ is more unstable in PEG toluene solution which leads to enzyme inactivation.
