青梅提取物防治氧嗪酸钾致小鼠高尿酸血症的研究 被引量:10

Study on the Prevention and Treatment of Prunus mume Extracts against Potassium Oxonate Induced Hyperuricemia in Mice

摘要 本文采用尿酸酶抑制剂氧嗪酸钾建立小鼠高尿酸血症模型,研究青梅核醇提物乙酸乙酯相(PMS-E)和青梅花醇提物正丁醇相(PF-B)对小鼠高尿酸血症的防治作用。小鼠血尿酸水平的测定采用碱性磷钨酸盐法,血清生化指标(肌酐、尿素氮和白球比)以及血清和肝脏中黄嘌呤氧化酶(XO)活性的测定采用试剂盒法。实验结果表明,PMS-E和PF-B在中、高剂量时均能显著降低高尿酸血症小鼠血尿酸水平(p<0.01);PF-B的降肌酐能力要强于PMS-E,但降尿素氮能力却弱于PMS-E;PMS-E和PF-B对血清白球比的影响与正常组比较均不显著(p>0.05)。PF-B的降肝脏XO能力要强于PMS-E,但在血清XO的降低方面两者差异不显著。本研究结果为青梅提取物用于开发降尿酸、抗痛风的功能性食品和膳食补充剂奠定了基础。 This paper mainly studied the prevention and treatment of ethyl acetate fi'action of Prunus mume seed extract (PMS-E) and n-butanol fraction of Prunus mume flower extract (PF-B) against potassium oxonate (uricase inhibitor) induced hyperuricemia in mice. The serum uric acid level of mice was measured by alkaline phosphotungstate method; serum biochemical indexes, such as creatinine, urea nitrogen and albumin and globulin ratio, and xanthine oxidase (XO) in serum and liver were measured by corresponding kit. The results indicated that the serum uric acid level in middle and high dose groups of PMS-E and PF-B significantly lower than hyperuricemic group (p〈0.01); PF-B and PMS-E could decrease the serum creatinine and urea nitrogen levels significantly, but had no significantly influence on albumin and globulin ratio 09〉0.05). In addition, PMS-E and PF-B could inhibit XO activity in serum and liver significantly. This study laid a foundation for Prunus mume extracts exploitation to functional foods and dietary supplements for lowering uric acid and anti-gout.
