抗抑郁药噻萘普汀钠的合成新工艺

A Novel Process for Antidepressant Tianeptine Sodium

摘要 根据文献报道的抗抑郁药噻萘普汀钠的合成方法,设计了一条新的合成路线。本论文以N-(3-氯-6,11-二氢-6-甲基-苯二氮卓[c,f][1,2]噻氮卓-11-酮)-S,S-二氧化物(5,简称三环酮)为起始原料,经还原、氯化、胺化、水解反应制得噻萘普汀钠(1),总收率为48.5%。产物结构经过IR和1HNMR确证。 A novel process for synthesis of tianeptine sodium was presented according to related literatures. Commencing with 3-chloro-6-methyl-dibenzo[c,f][1,2]thiazepin-11(6H)-one-5,5-dioxide(5), tianeptine sodium was produced in four steps of reduction, chlorination, amination and hydrolysis in 48.5 % total yield. The structure was confirmed by 1R and IHNMR. The novel process fit industrial production very well.
