摘要
目的探讨依托泊苷联合顺铂对骨肉瘤细胞增殖和凋亡的影响,为骨肉瘤的治疗找寻新的化疗方法。方法采用MTT法检测药物对骨肉瘤细胞增殖的影响。倒置相差显微镜来观察用药前后细胞形态改变,用流式细胞仪检测药物分别及联合作用骨肉瘤细胞后细胞凋亡情况。结果依托泊苷联合顺铂能明显抑制骨肉瘤细胞的生长,与两药单独使用相比较,联合用药抑制骨肉瘤细胞增殖的作用明显增强。依托泊苷联合顺铂作用于骨肉瘤细胞后凋亡率增高。倒置相差显微镜可见两药联用时大量骨肉瘤细胞凋亡脱落。结论依托泊苷联合顺铂能有效抑制骨肉瘤细胞增殖,且促进骨肉瘤细胞凋亡,具有协同抗肿瘤作用。
Objective To investigate the influence of combination of etoposide and cisplatin on proliferation and apoptosis of osteosareoma cells,to find a new method for treatment of osteosarcoma. Methods MTT method was used to detect the influence of the drug on the proliferation of the osteosarcoma cell.Inverted phase contrast microscope was used to observe the change of cell shape before and after drug intervention,flow cytometry was used to detect effects of osteosarcoma cell apoptosis pharmacy separately and combined. Results The combination of etoposide and cisplatin could inhibit the growth of osteosarcoma cells,the combination of etoposide and cisplatin increases rates of the apoptosis of osteosarcoma cell and inhibits proliferation of osteosarcoma cell.Apoptosis and shedding of osteosarcoma cells were visible on inverted phase contrast microscope. Conclusion The combination of etoposide and cisplatin could effectively inhibit the proliferation of osteosarcoma cells, promote apoptosis of osteosarcoma cells,and has a synergistic antitumor effect.
