摘要 MYB转录因子广泛参与植物的生长、发育、生理代谢的调控及胁迫应答,它们均具有保守的MYB结构域。GmMYBJ7是从大豆中分离获得的一个新的MYB基因(GenBank登录号:DQ902864)。亚细胞定位检测结果表明,Gm-MYBJ7蛋白定位于细胞核中;半定量RT-PCR检测结果显示,在紫外辐射、高盐及干旱(PEG)等非生物胁迫处理下,GmMYBJ7在大豆品种吉林3号中的表达量明显降低;构建植物表达载体pCAMBIA2301-GmMYBJ7,利用农杆菌介导法转化烟草NC89,在GmMYBJ7的阳性转化受体中,总黄酮的含量则明显减少。推测GmMYBJ7可能通过对类黄酮生物合成的调控参与植物的基础生理代谢。 Plant MYB transcription factors are characterized by the presence of a structurally conserved MYB domain,which involved in the regulation of many aspects of plant growth,development,metabolism and stress responses.A new MYB gene,designated GmMYBJ7(GenBank accession No.DQ902864),was isolated from soybean(Glycine max).The result of subcellular localization indicated that GmMYBJ7 proteins were mostly distributed in the nucleus.Semi-quantitative RT-PCR analysis indicated that the expression of GmMYBJ7 would be decreased under the abiotic stresse treatment such as UV-B radiation,drought,and high-salt.GmMYBJ7 were transformed into tobacco NC89 with Agrobacterium LBA4404 containing the plant expression vectors pCAMBIA2301-GmMYBJ7.The results showed that the average total flavonoids content decreased significantly in the positive tobacco transformants,which suggested that GmMYBJ7 was probably invoved in the basic physiological metabolism by regulating the biosynthesis of flavonoid in plants.
