利用 D2 5 4 树脂可以将原始粗品肝素钠的效价提高到 179.8U SP U/m g,进一步纯化可得 2 0 1.5U SP U/m g的精品 ,其 H2 O2 用量 ( 1.5% )和氧化时间 ( 2 4 h)均少于传统工艺 ,产品光吸收和收率 ( 85.8% )
A procedure for purifying sodium heparin from its crude preparation was developed. Heparin was adsorbed on resin D 254 under the condition of pH4.0 and 1.5 mole/1 NaCl and was eluted with 3.5 mole/1 NaCl. Heparin in the eluate was precipitated with 45% ethanol. Valence of heparin at this step was increased to 179.8 USP μ/mg. After removing acidic protein with precipitator YNB 99 1 and basic protein with the oxidization of 1.5% H 2O 2 for 24 hours, the valence of hepariu was increased to 201.5 USP μ/mg. Comparing with traditional process, the quality of heparin produced with the new method was the best and the yield was the highest (85.8%).
