解偶联蛋白3基因多态性与中国人2型糖尿病的相关性研究 被引量:2

摘要 目的 探讨解偶联蛋白 3( U CP3)基因二核苷酸 ( CA) n串联重复序列多态标记与中国汉族人 2型糖尿病的关系。方法 用 PCR-变性聚丙烯酰胺凝胶电泳及银染方法 ,检测 2 0 8例南京地区汉族人 ( 2型糖尿病 10 6例 ,正常对照 10 2例 )U CP3基因的 ( CA) n串联重复序列多态标记 ,比较各组间的等位基因频率和基因型频率。结果  2型糖尿病组和正常对照组U CP3基因等位基因频率的分布存在显著性差异 ( χ2 =2 6 .12 ,P<0 .0 0 5 ) ;2型糖尿病组 UCP36、7、8等位基因频率显著高于正常对照组 ( RR=4.49,P<0 .0 0 5 ;RR=7.14,P<0 .0 0 5 ;RR=14.2 3,P<0 .0 5 ) ;中国南京地区汉族人群与高加索人群 UCP3基因等位基因频率的分布亦有显著性差异 (χ2 =37.43,P<0 .0 0 5 )。结论  U CP3基因 ( CA) n串联重复序列多态标记与中国汉族人 Objective To investigate the association between dinucleotide (CA)n tandem repeat polymorphism of uncoupling protein 3 gene and type 2 diabetes mellitus in Chinese. Methods A casecontrol study on 208 Chinese subjects from Nanjing region (106 patients with type 2 diabetes and 102 normal subjects) was performed. The polymorphism of (CA)n tandem repeat of UCP 3 gene was determined by PCRdenaturing polyacrylamide gel electrophoresis and silver staining. Results The prevalence of UCP 3 gene allele frequency in type 2 diabetic patients was significantly different from that in control subjects. An elevated frequency of 6,7,8 allele was observed in type 2 diabetes mellitus as compared with control subjects. Significant differences of UCP 3 gene allele frequency were found between the Chinese and the Caucasian population. Conclusion Dinucleotide (CA)n tandem repeat polymorphism of uncoupling protein 3 gene is associated with type 2 diabetes mellitus in Chinese.
