Ⅰ型胶原及其受体系统在成骨细胞内的表达 被引量:13

EXPRESSION OF TYPE Ⅰ COLLAGEN AND ITS RECEPTOR SYSTEM IN OSTEOBLASTS

摘要 目的 研究 型胶原及其受体系统在不同代次成骨细胞内的表达情况。方法 选用原代、第 6代及第 15代人胚骨膜成骨细胞 ,S- P免疫组织化学染色观察 型胶原及整合素α2 β1 的表达情况 ,定量 RT- PCR技术检测 型胶原及整合素α2 β1 的 m RNA表达情况。结果  型胶原及其受体在不同代次成骨细胞内均有表达 ,免疫组织化学未发现各代次间表达量的差异 ;不同代次的人胚成骨细胞都有 型胶原及其受体 -整合素α2 β1 的m RNA表达 ,表达量随成骨细胞传代次数的增加逐渐减弱。结论  型胶原是通过整合素受体α2 β1 对成骨细胞发挥促分化和促粘附作用 ; Objective To study the expression of type Ⅰ collagen and its receptor system integrin α 2β 1 in different passages of osteoblasts.Methods The expression of type Ⅰ collagen and integrin α 2β 1 in the primary, sixth and fifteenth passage of osteoblasts were detected by S P immunohistological staining technique, and their mRNA expression by quantity RT PCR technique.Results Type Ⅰ collagen and integrin α 2β 1 were expressed in different passages of osteoblasts and there was no significant difference among three passages by immunohistological technique. Their mRNA expression was gradually decreased with subculture.Conclusion Type Ⅰ collagen promotes the adhesion and phenotype expression of osteoblasts through its receptor integrin α 2β 1. The reductive expression of type Ⅰ collagen receptor system will decline the phenotype of osteoblasts.
