摘要
目 的 观察人类甲状腺癌中c -erbB - 2癌蛋白和mRNA的表达及其关系。方法 应用免疫组化和同位素点杂交技术对新鲜冰冻甲状腺组织c -erbB - 2癌蛋白和mRNA的表达进行研究。结果 2 1例甲状腺乳头状癌中 14例c -erbB - 2癌蛋白表达阳性 ,而其他类型肿瘤和非肿瘤组织均未见c -erbB -2表达。c -erbB - 2mRNA表达研究显示 ,乳头状癌和淋巴结转移灶中mRNA的表达水平明显高于其他类型肿瘤和非肿瘤组织。本研究还证明石蜡包埋的乳头状癌标本中c -erbB - 2癌蛋白表达率明显低于新鲜标本。结论 c -erbB - 2蛋白的表达是甲状腺乳头状癌的一个特征性改变 ,c -erbB - 2蛋白的表达与mR NA的表达相一致 ,c -erbB - 2蛋白检测的稳定性取决于组织标本的质量。
Objective To investigate the expression and relationship of c-erbB-2 oncoprotein and mRNA in human thyroid carcinomas. Methods c-erbB-2 oncoprotein and mRNA were detected in frozen thyroid tissue specimens using immunohistochemistry and isotope Dot blot hybridisation. Results c-erbB-2 immunoreactivity was detected in 14 out of 21 papillary carcinomas, while no c-erbB-2 oncoprotein immunstaining was seen in cases of the other thyroid tumors and non-neoplastic tissue. C-erbB-2 mRNA was analysed by Dot blot hybridisation. Papillary carcinomas and lymph node metastases showed significantly higher levels of c-erbB-2 mRNA compared to the other thyroid tumors and non-neoplastic tissue. The positive rate of c-erbB-2 oncoprotein on formalin-fixed, paraffin-embedded material were much lower than levels on frozen tissue. Conclusion The present work presents c-erbB-2 oncoprotein expression as a feature of papillary thyroid carcinomas, and is consistent with mRNA expression. The detecting stability of c-erbB-2 is depand on optimal tissue quality.
