摘要
目的利用软件预测羊口疮病毒(ORFV)F1L蛋白的二级结构和细胞表位。方法利用SOPMA服务器预测ORFV F1L蛋白的二级结构,以DNAStar软件单参数(二级结构、亲水性、可及性、柔韧性及抗原性)预测结果为基础,通过二级结构预测初步筛选,并以ABCpred方案作为最终验证,预测羊口疮病毒(ORFV)F1L蛋白的B细胞表位;运用神经网络+量化矩阵法(ANNs+QM法)预测ORFV F1L蛋白的CTL细胞表位;使用MHC-II类分子结合肽在线程序预测ORFV F1L蛋白的Th细胞表位。结果 ORFV F1L蛋白含有α-螺旋34.71%、β-片层18.82%、β-转角5.88%、无规则卷曲40.59%;没有信号肽,可能存在7个B细胞表位,2个CTL表位,3个Th细胞表位。结论该研究结果将为建立ORFV诊断方法、制备单克隆抗体及合成肽疫苗提供理论依据。
The network platform SOPMA server was used to predict the secondary structure of ORFV F1L protein.On the basis of the DNAStar prediction results(hydrophilicity,accessibility,flexibility and antigenicity),the B cell epitopes of ORFV F1L protein was predicted by screening the secondary structure and followed by verification using ABCpred program.The cytotoxic T cell(CTL) epitopes of ORFV F1L protein were predicted by combination of quantitative matrix(QM) and artificial neural network(ANN).T helper(Th) epitopes of ORFV F1L protein were predicted by an online server for the prediction of MHC class II-peptide binding affinity.The result showed that ORFV F1L protein had alpha helixes(34.71%),random coils(40.59%),β-sheet(18.82%) and β-turn(5.88%).No signal peptide was found,while 7 B-cell epitopes,2 CTL epitopes and 3 Th epitopes of ORFV F1L protein were predicted.These findings would provide theoretical basis for establishment of ORFV diagnosis method,preparation of monoclonal antibody and development of vaccine.
