摘要
目的探讨阿托伐他汀对糖尿病肾病大鼠肾小球细胞增殖的影响及意义。方法将40只SD雌性大鼠随机分为四组:糖尿病肾病组(DN组)、预防组、治疗组、对照组,每组10只。DN组、预防组和治疗组糖尿病造模成功后,预防组和治疗组分别给予不同剂量的阿托伐他汀干预,对照组仅注射柠檬酸缓冲液。第8周测定相关生化指标,观察肾组织病理改变后,通过免疫组化法分析大鼠肾小球增殖细胞核抗原(proliferation cell nuclear antigen,PCNA)的表达。结果预防组和治疗组相关生化指标较DN组好转,肾脏病变减轻,PCNA表达于肾小球固有细胞。DN组肾小球中PCNA的表达均较预防组、治疗组、对照组明显上调。而预防组、治疗组肾小球中PCNA的表达较DN组下调,且预防组PCNA的下调更为显著。结论阿托伐他汀通过下调肾小球PCNA表达,抑制肾小球细胞增殖实现对DN的保护作用。
Objective To study the effect and significance of atorvastatin on glomenclar cell proliferation in rats with diabetic nephropathy. Methods 40 female SD rats were randomly divided into four groups: the diabetic nephrop- athy group (DN group), the prevention group, the treatment group and the control group, with 10 cases in each group. After the diabetic rat models of the DN group,the prevention group and the treatment group were being made successfully, the prevention group and the treatment group were treated with different doses of atorvastatin. The con trol group were only injected with citric acid buffer. After detecting related biochemical indexes and observing the pathological changes of renal tissue in the eighth week, expression of proliferation cell nuclear antigen(PCNA)in rat glomerular were analysed by means of immunohistochemistry. Results The prevention group and the treatment group had Better improvement in their biochemical indexes than the DN group and also had attenuated lesions in renal tissue. The expression of PCNA in glomerular ceils in the DN group were significantly increased compared to the prevention group,the treatment group and the control group. However, the expression of PCNA in glomerular of the prevention group and the treatment group were on down regulation, and the down-regulation in the prevention group was more significantly obvious. Conclusion Atorvastatin has protective effect on DN by reducing the expression of glomerular PCNA and inhibiting the proliferation of glomerular cells.
