rhBMP-2/20%-β-TCP/SF复合物植入兔股骨缺损的实验研究

Experimental Study on the Reconstruction of Thighbone Defects with rhBMP-2/20%-β-TCP/SF Composite in Rabbits

摘要 目的探讨rhBMP-2/20%-β-TCP/SF复合物的生物相容性及其作为骨替代材料的可行性。方法将12只健康雄性新西兰兔随机分为实验组和对照组,每组6只,按Thomas方法,在其左侧股骨内侧髁作9 mm×4 mm×5 mm的骨缺损区模型,然后将rhBMP-2/20%-β-TCP/SF复合物材料植入实验组,对照组植入自体骨碎片。分别于术后4周、8周、12周各处死2只动物模型进行大体观察,以及进行X线、人工骨降解情况及早期骨组织学切片分析。结果术后12周实验组植入材料基本完全溶解,并形成典型的骨小梁及髓腔结构,成骨面积为50.45%,大于对照组的42.17%。结论 rhBMP-2/20%-β-TCP/SF复合物具有良好的生物相容性,能有效促进骨缺损的修复愈合,有望成为修补股骨缺损的人工骨移植材料。 Objective To explore the biocompatibility of rhBMP-2/20%-β-TCP/SF composite and the feasibility of applying the composite to bone implantation. Methods Twelve male rabbits were divided into two groups in the study: experimental group (n =6 ) and control group (n =6 ). Bone defects of 9 mm × 5 mm ×4 mm were made at the entocondyle of left side of thighbone in rabbits according to Thomas Method. rhBMP-2/20%-β-TCP/SF composite was used as the implant in the experimental group,while autologous bone piece was used in the control group. Two animals were killed at 4 weeks ,8 weeks and 12 weeks after operation for gross observation,X-my observation,artificial bone degradation and early histological manifestations were done respectively. Results Twelve weeks after operation,rhBMP-2/20%-β-TCP/SF composite was almost degraded in the experimental group ;the trabecular bone and canal structure were also formed in experimental group. The osteogenesis area of experimental group was larger than that of control group(50. 45% vs 42. 17% ). Condusion rhBMP-2/20%-β-TCP/SF composite is a kind of artificial bone substitute material with good biocompatibility, which can effectively accelerate the healing of bone defects.
