摘要
目的:探讨苗药皮部熏洗法治疗骨关节炎的部分机理。方法:取42只健康清洁级大鼠,随机选取8只不造模作为正常对照组,其余34只采用4%木瓜蛋白酶关节腔注射建立大鼠OA模型,1周后随机选取2只检测模型成功与否,将造模成功的32只大鼠随机分为模型组、熏组、洗组和熏洗组,治疗4周后处死动物,切取关节,免疫组化染色法测定p53、bcl-2的表达。结果:正常组大多数标本未见p53与bcl-2表达,造模后见到散在的p53与bcl-2表达,熏洗后bcl-2的表达有逐渐增强、p53的表达有逐渐减弱的趋势。结论 (:1)通过苗药皮部熏洗防治的干预,对OA模型大鼠的痛行为及膝关节活动有明显的改善作用。(2)苗药皮部熏洗通过上调软骨bcl-2的表达和下调p53的表达,来延缓软骨细胞的凋亡。(3)熏洗法治疗大鼠OA比单独运用熏法、洗法效果更明显。
Objective：To investigate the mechanism of the Miao medicine,fumigation method to treat osteoarthritis.Methods：42 healthy and clean rats were randomly selected,eight non-modeling as a normal control group,the remaining 34 were given 4% papain intra-articular injection to make rat OA model,and a week later 2 were randomly selected to test the model success,the 32 model rats were randomly divided into model group,smoked group,wash group and fumigation group,animals were sacrificed after 4 weeks of treatment,immunohistochemical staining method for the determination of p53 and bcl-2 expressions.Results：The majority of specimens of the normal group had no expression of p53 and expression of bcl-2,after the model,there were scattered p53 and bcl-2 expressions,fumigation made bcl-2 expression gradually increased,p53 expression gradually weakened.Conclusion：（1）Through the intervention of the Miao medicine prevention and treatment,pain behavior and knee OA symptorns were significantly improved.（2）Fumigation with Miao medicine raised the cartilage bcl-2 expression and decreased of p53 expression to delay the apoptosis of chondrocytes.（3）Fumigation treatment of rat OA is more obvious than the use of single washing method effect.
