凤尾草总黄酮抑制骨肉瘤MG-63细胞迁移及其机制的初步研究 被引量:4

Effect of the Pteris Multifida Flavones on the Metastasis Ability of Human Osteosarcoma Cell Line MG-63 Cells

摘要 目的:观察凤尾草总黄酮对人骨肉瘤MG-63细胞体外迁移能力的影响,并探讨其作用机制。方法:利用乙醇萃取法制备凤尾草总黄酮,通过划痕实验、Transwell实验观察不同浓度凤尾草总黄酮对同样条件培养下骨肉瘤MG-63细胞迁移能力的影响,并采用Western blot方法检测不同浓度凤尾草总黄酮对MG-63细胞内白介素6(IL-6)蛋白表达水平的影响。结果:同未加药物对照组相比,经高浓度凤尾草总黄酮(5与10 g.L-1)处理后,MG-63细胞的划痕愈合能力明显减弱,穿膜细胞的数量显著减少;同时,细胞内IL-6蛋白的表达水平明显降低,并与凤尾草总黄酮的浓度呈负相关性。结论:凤尾草总黄酮抑制了骨肉瘤MG-63细胞的迁移能力,其作用与抑制MG-63细胞内的IL-6蛋白表达水平有关。 Objective:To investigate the effect of the Pteris multifida Flavones on MG-63 cells metastasis of human osteosarcoma cell line in vitro and to explore the action mechanisms of the Pteris multifida Flavones.Methods:Pteris multifida preparation was made by ehanol,the effect of the different concentrations of Pteris multifida Flavoneson the experimental metastasis ability of MG-63 cells was observed by scratch test and Transwell metastasis experiments.The levels of IL-6 protein in MG-63 cells were detected by Western Blotting.Results:Comparing with control group,the Pteris multifida Flavones(5 and10 g·L-1)inhibited the MG-63 cells wound closure significantly at 24 hours time points and decreased the number of cells passing through filter membrane.The levels of IL-6 protein in MG-63 cells were decreased significantly after treated with the Pteris multifida Flavones.Conclusion:The Pteris multifida Flavones is able to inhibit the metastasis ability of MG-63 cells.The action mechanisms of the Pteris multifida Flavones are related to the inhibition of IL-6 protein levels in MG-63 cells.
