摘要
上皮生长因子(EGF)属于一类激素样细咆调节因子,可促进多种正常细胞及肿瘤细胞的体外增殖。上皮生长因子受体(EGFR)是一分子量为170 000—180 000道尔顿的细胞表面多肽,其组织学分布较广泛,在某些肿瘤细胞上有异常高的表达。如源自女性外阴部上皮癌的A431细胞和源自人乳腺癌的MDA-468。本研究报告了在源自人鼻咽部低分化鳞癌的细胞株CNE-2 Z上有较高的EGFR表达(1.3×10 10~5/细胞)。当EGF终浓度低予1×10^(-10)M,对A431细胞、CNF-2 Z细胞的体外增殖有着和缓的促进作用;当EGF的终浓度为1×10^(-9)M时,A431细胞和CNE细胞分别受抑制,而CNE-2 Z细胞仍处于促增殖状态;随着EGF浓度的进一步增高,CNE-2 Z亦转为受抑状态;当EGF浓度为1×10^(-7)M时,A431和CNE完全受抑制;CNE-2Z抑制率为61％。以上结果提示高浓度的EGF对CNE-2 Z的体外增殖具有抑制作用。
Epidermal growth factor (EGF) , a hormone-like cytomodulatory fact-tor, has been noted to stimulate the prolliferation of a variety of normal and malignant cells, We report here that CNE-2Z, a human poorly differentiated nasopharyngeal carcinoma cell line with a high number of EGF receptors(1.3× 10 5 /cell), is growth inhibited by EGF. The growth of CNE-2Z cells was mildly stimuiated by EGF under concentrations of 10-9 to 10-11 M and was significantly inhibited by EGF at concentrations greater than10-8 M, while, under the serum containing conditions employed, A431 cell line was growth inhibit, ed by EGF at concentration of 10-9M The results suggest that CNE- 2Z is growth inhibited by high concepntration of EGF and the inhibitory effect may be not independent on the number of EGF receptors.
