摘要
目的:总结国内外对5-氟尿嘧啶(5-FU)类药物抗癌新调控机制的最新进展,为氟尿嘧啶类药物的临床应用以及个体化用药提供依据。方法:应用Medline、PubMed及CNKI数据库检索系统,以"氟尿嘧啶、细胞凋亡、活性氧和DNA错配修复"等为关键词,检索2005-11-2012-01的相关文献,共检索相关文献26篇。纳入标准:1)5-FU类药物诱导肿瘤细胞凋亡作用;2)5-FU类药物通过活性氧(ROS)途径促进细胞凋亡;3)错配修复(MMR)系统及其他分子如P450R、PKC-δ等因素对5-FU类药物抗癌作用的调控。根据纳入标准符合分析的文献26篇。结果:除经典的抗癌作用机制外,5-FU类药物通过死亡受体途径和线粒体途径诱导细胞凋亡,还可以通过增加ROS含量促进细胞凋亡,同时抗癌作用受到MMR系统和P450R等因素的调控。结论:对5-FU类药物的抗癌作用机制及调控因素的研究,有助于为临床指导用药提供理论依据。
OBJECTIVE:To summarize the latest development of the novel regulating on anticancer effects of 5-flu- orouracils in domestic and abroad, and provide the basis for the clinical application and individualized medicine of 5-flu- orouraeils. METHODS: “fluorouracil,apoptosis,reactive oxygen species(ROS) and DNA mismatch repair” were used as keywords to retrieval the literature from the database retrieval systems of Medline, PubMed and CNKI from November 2005 to January 2012. Including standards:1) apoptosis induction of 5-fluorouracils; 2) apoptosis induction of 5-fluoroura- cils through the way of ROS; 3) the influence factors of MMR and P450R involved in 5-fluorouracil's anticancer regula- ting mechanism. And 26 papers were selected and analyzed. RESULTS: Beside the classic anti-cancer effect mechanisms, 5-fluorouracils induce cells apoptosis through death receptor pathway and mitochnodria pathway,and promote cell apopto- sis through increasing ROS,and then the anticancer effects are regulated by MMR,P450R and other molecules. CONCLU- SION:The research of anticancer effects and regulating factors of 5-fluorouracils can provide a theory basis for guiding clinical medication.
