左甲状腺素和维生素E对甲状腺功能减退症大鼠心肌损伤的影响 被引量:7

摘要 目的研究左甲状腺素(L-T4)、维生素E(VitE)及两者联合治疗甲状腺功能减退症(简称甲减)大鼠,了解其对大鼠心肌酶谱、心肌病理及氧化应激水平的影响,探讨甲减心肌的治疗方法。方法将SD雄性大鼠随机分为正常对照组、模型组、T4治疗组、VitE治疗组、T4+VitE联合治疗组。测定血清三碘甲腺原氨酸(T3)、甲状腺激素(T4)、促甲状腺激素(TSH)水平,血清心肌酶谱[肌酸激酶(CK)、肌酸激酶同工酶(CKMB)、乳酸脱氢酶(LDH)、谷草转氨酶(AST)]水平,各组心肌组织HE染色观察心肌细胞形态,测定血清及心肌组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果①与正常对照组比较,模型组、VitE治疗组T3、T4水平明显降低,TSH显著升高(P<0.05);与模型组比较,T4治疗组、T4+VitE联合治疗组T3、T4水平明显升高,TSH显著降低(P<0.05);模型组与VitE治疗组比较T3、T4、TSH水平差异无统计学意义。②与正常对照组比较,模型组血清CK、LDH、AST水平显著升高(P<0.05);与模型组比较,T4治疗组、VitE治疗组、T4+VitE联合治疗组的CK、LDH、AST水平显著降低(P<0.05),CKMB差异无统计学意义。③血清SOD与LDH、AST、CK、CKMB呈显著相关性。④HE染色镜下观察:与正常对照组比较,模型组心肌可见黏液性变等病理改变;与模型组比较,T4治疗组、VitE治疗组、T4+VitE联合治疗组心肌均有改观。⑤与正常对照组比较,模型组血清及心肌组织MDA含量显著升高(P<0.05);与正常对照组比较,模型组血清SOD活性显著降低(P<0.05),但各组间心肌组织SOD活性差异无统计学意义。结论 T4、VitE及联合治疗能明显降低甲减大鼠心肌酶谱水平,改善心肌细胞病理状态,可能与甲状腺功能改善和氧化应激水平降低有关。 Objective To investigate the myocardial enzymes,myocardial pathology and oxidative stress status in propylthiouracil-induced hypothyroidism rats and examine the effect of levothyroxine,vitamin E(VitE) and both supplementation on this experimental model.Methods Male Sprague Dawley rats were randomly divided into normal control group,model group,T4 treatment group,VitE treatment group,T4+VitE combination therapy group.Serum T3,T4,TSH levels,serum myocardial enzymes,serum and myocardial superoxide dismutase(SOD) activity and malondialdehyde(MDA) content were measured.To contrast each group of myocardial cell morphology through hematoxylin and eosin(HE) staining.Results ① Compared with normal control group,T3 and T4 levels were significantly decreased and TSH level was significantly elevated in model group and VitE treatment group(P0.05).Compared with model group,T3 and T4 levels were significantly elevated and TSH level was significantly decreased in T4 treatment group and T4+VitE combination therapy group(P0.05).There was no statistical difference in T3,T4 and TSH levels between model group and VitE treatment group.② Compared with normal control group,CK,LDH and AST levels were significantly elevated in model group(P0.05).Compared with model group,they were significantly decreased in T4 treatment group,VitE treatment group and T4+VitE combination therapy group(P0.05),the CKMB difference was not statistically significant.③ Serum SOD and LDH,AST,CK,CKMB was a significant correlation.④ HE stain microscope observation showed that compared with control group,there were some mucous changes in myocardium of model group.Compared with model group,the condition of myocardial injury improved in T4 treatment,VitE treatment and T4+VitE treatment group.⑤ Compared with control group,serum and myocardial MDA levels in model group increased significantly(P0.05),serum SOD activity in model group decreased significantly(P0.05),between myocardial SOD activity in each group the difference was not statistically significant.Conclusion Levothyroxine,VitE and both treatment of hypothyroidism in rats can significantly reduce the myocardial enzymes,improve the pathology conditions,which may be related to the improvement of thyroid function and reduction of the level of oxidative stress.
