苦豆子总碱对大鼠胃窦环形平滑肌作用机制的研究 被引量:5

Researching the Mechanism of Action on Total Alkali Sophora alopecuroide L to Sinus Ventriculi Circular Muscle in Vitro of Rats

摘要 目的:观察苦豆子总碱(total alkali sophora alopecuroids L,TASa)对大鼠胃窦环形肌条的作用及机制。方法:采用离体恒温灌流浴槽,记录胃窦环形肌条收缩波的振幅、持续时间、收缩面积和频率,给予TASa(5、10、20、40、80mg/L)观察其量效关系(r);将标本分为对照组(NS)、TASa、TASa+阿托品(10-6mg/L)、TASa+酚妥拉明(10-6 mg/L)、TASa+苯海拉明(10-6 mg/L)、TASa+消炎痛(10-6 mg/L)和TASa+异搏定(10-7 mg/L)七组探讨TA-Sa收缩肌条的作用机制。结果:TASa能增强肌条收缩的振幅、持续时间和收缩面积,并有量效依赖性(振幅:r=0.96,t=23.72,P<0.01;持续时间:r=0.95,t=32.55,P<0.01;收缩面积:r=0.98,t=23.68,P<0.01),但对其收缩频率(NS:3.58±0.15,TASa:3.72±0.21,P>0.05)无影响;异搏定能明显抑制TASa收缩肌条的作用,而阿托品、酚妥拉明、苯海拉明和消炎痛无影响。结论:TASa可能是通过激活Ca2+通道,升高胞浆内Ca2+而加强肌条的收缩。 Objective: To study the contractive effects of the total alkali sophora alopecuroids L (TASa) on sinus ventriculi circular muscle in vitro of rats and the possible mechanisms. Methods : Adopted constant temperature perfusion of the bath ma- chine, record amplitude, duration, contraction area, frequency of the contraction sinus ventrieuli circular muscle in vitro, TA- Sa divided into five doses groups ( 5, 10, 20, 40, 80mg·L-1 ), giving different doses of TASa to sinus ventrieuli circular mus- cle in vitro and observed the dosc-effect relationship between muscle contraction and different doses of TASa. Sinus ventriculi cir- cular muscle were divided into seven groups, including control group, TASa group, TASa+10^-6 mg/L atropine group, TASa+ 10^-6 mg/L phentolamine group, TASa+ 10^-6 mg/L benzhydramine group, TASa+ 10^-6 mg/L antifani group, TASa+ 10^-7 mg/L isop- tin group. The contraction effects of TASa were investigated. Results: TASa could increase contraction amplitude, contraction duration, contraction area, and has an obvious concentration-dependent contraction of sinus ventriculi circular muscle ( contrac- tion amplitude: r=0.96, t=23.72, P〈0.01; contraction duration: r=0.95, t=32.55, P〈0.01; contraction area: r= 0. 98, t=23. 68, P〈0. 01 ), but had no effect on contraction frequency (control group: 3.58±0. 15, TASa: 3.72±0. 21, P〉 0. 05 ). Isoptin could inhibit the circular muscle contraction of TASa, atropine, phentolamine, benzhydramine and antifani had no contraction effect. Conclusion: TASa may activate calcium channel, increase calcium flow to the interior of the cell and in- crease the contraction of sinus ventrieuli circular muscle.
