摘要
利用35个SSR标记,分析了巨桉(Eucalyptus grandis)4个种源的遗传变异,鉴定了与枝瘿姬小蜂(Leptocybeinvasa)抗性相关的SSR位点。共检测到469个等位片段,平均每位点的等位片段数(NA)、观测杂合度(Ho)、期望杂合度(He)和多态性信息量(PIC)分别为13.3、0.5385、0.8101和0.7797,供试群体的遗传多样性较高;分子方差分析(AMOVA)表明,种源内个体间变异占96.0%,种源间变异占4.0%,这与群体间较低的分化系数(Fst=0.0623)和较高的基因流(Nm=4.09~14.13)一致。聚类分析表明,种源14393与种源15358的亲缘关系最近(Fst=0.017),种源14393与种源20261最远(Fst=0.058)。关联分析检测到EST-SSR226与枝瘿姬小蜂抗性显著相关(P=0.03),对抗性的决定系数达0.4436,增效效应为0.71,增效比例42.8%。
Thirty-five simple sequence repeat (SSR) markers were used to assess genetic diversity of four Eucalyptus grandis provenances and identify markers significantly associated with resistance to gall wasp (Leptocybe invasa) in E. grandis. A total of 469 alleles were produced with the 35 SSRs, averaging at 13.3 alleles per marker. The mean values of observed heterozygosity (Ho), observed heterozygosity (He) and polymorphic information content (PIC) were 0.5385, 0.8101 and 0.7797, respectively, across all the SSR loci, indicating a high level of genetic diversity. AMOVA analysis revealed that genetic variation among individuals within provenances accounted for 96.0% of the total molecular variance and 4.0% of the variance occurred among provenances, which coincided well the low Fst (0.0623) and high Nm (4.09 - 14.13). UPGMA cluster analysis shows that provenances 14393 and 15358 were the most close (Fst=0.017) whereas provenances 14393 and 20261 were the most distant (Fst=0.058). The marker EST-SSR226 was identified to associate with resistance to gall wasp (P=0.03), with a determination coefficient 0.4436, positive effect 0.71 and positive effect percentage 42.8%.
