摘要
本文用自制 18-羟皮质醇抗血清 ,建立血浆 18-羟皮质醇竞争性 ABC- EL ISA。同时测定部分正常人、原发性醛固酮增多症 (原醛 )和原发性高血压病人血浆 18-羟皮质醇水平。结果该方法批内变异为 6 .6 %～ 8.1% ,批间变异为 11.3%～ 12 .2 % ,平均回收率为 10 2 .1% ,最低可测定限为 0 .42 nmol.L- 1 ,正常参考值为 3.0± 1.7nmol.L- 1。原醛腺瘤组血浆 18-羟皮质醇明显高于原醛增生组 ,原发性高血压组和正常组 (P<0 .0 5 ) ,后三组之间无显著性差异 (P>0 .0 5 )。鉴于本法具有简便、灵敏、稳定等特点 。
The antisera of 18 hydroxycortisol were produced by immunizing rabbit and a competitive avidin biotin enzyme linked immunoassay (ABC ELISA) for plasma 18 hydroxycortisol was established. Using the assay, we measured the levels of 18 hydroxycortisol in normal persons, patients with primary aldosteronism and essential hypertension. Mean rate of recovery of this assay was 102 1%,the intra and inter assay of coefficients of variation were 6 6%～8 1% and 11 3%～12 2%,respectively. The detection limit of plasma was 0 42nmol.L -1 .The normal value of 18 hydroxycortisol was 3 0±1 7nmol.L -1 . the levels of 18 hydroxycortisol in patients with aldosterone producing adenoma are significantly higher than that of other groups patients(P<0 05).Except the APA group,the levels of 18 hydroxycortisol in other groups had no significant difference(P>0 05).It could be used as a high sensitive and specifically discriminator for the differential diagnosis of primary aldosteronism.
