摘要
目的探讨蕨麻醇提取物对硫代硫酸钠(Na2S2O4)诱发的人神经母细胞瘤(SH-SY5Y)细胞株缺氧损伤的拮抗作用。方法取处于生长对数期的SH-SY5Y细胞,分别设空白对照(D-Hank液)组、缺氧模型(1 mmol/L连二亚硫酸钠)组及1、0.25、0.062 5 g/L蕨麻醇提取物拮抗缺氧(1 mmol/L连二亚硫酸钠)组,于5%CO2、37℃孵育4 h后,采用MTT试验检测细胞活性,测定上清液乳酸脱氢酶(LDH)活力,采用悬浮芯片蛋白法测定细胞内单核细胞趋化因子-1(MCP-1)、白细胞介素-1β(IL-1β)、IL-6、IL-10、γ-干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)、血管内皮生长因子(VEGF)水平7种细胞因子水平。结果与空白对照组比较,缺氧模型组和各剂量蕨麻醇提取物拮抗缺氧组SH-SY5Y细胞活性下降,LDH外漏量和空泡细胞率增大,MCP-1、IL-1β、IFN-γ和TNF-α水平升高,VEGF、IL-6和IL-10含量下降。与缺氧模型组比较,各剂量蕨麻醇提取物拮抗缺氧组细胞活性升高,LDH外漏量和空泡细胞率减少,细胞因子水平发生逆转,其中,MCP-1、IL-6、IFN-γ的改变以1 g/L组最为明显,IL-1β、IL-10、TNF-α、VEGF的变化以0.062 5 g/L组明显。结论蕨麻醇提取物可拮抗连二亚硫酸钠所致SH-SY5Y细胞缺氧损伤效应。
Objective To investigate the antagonistic effects of n-butanol extract of PotentiUa anserine L. on the hypoxia human neuroblastoma cell SH-SY5Y caused by sodium dithionite (Na2S2O4). Methods The exponential phase of SH-SY5Y cells were collected, respectively divided into control group,hypoxia group (exposed to 1 mmol/L Na2S2O4) and hypoxia potentilla groups. The hypoxia potentilla groups were treated with n-butanol extract of Potentilla anserine L. at different concentrations (1,0.25 and 0.062 5 g/L) and hypoxia, which were incubated at 37 ℃ under the condition of 5% CO2 for 4 h. The survival rate was detected by MTT assay,and the release quantity outward cell was detected by the lactate dehydrogenase (LDH) content. There were seven kinds of cytokine levels detected by suspended chip array technique,including monocyte chemotactic factor-1 (MCP-I), interleukin-1β (II,-113), IL-6, IL-10, γ-interferon (IFN-γ), tumor necrosis factor-α (TNF-α)and vascular endothelial growth factor (VEGF). Results Compared with the blank control group, the cell aet;vities of hypoxia group and each dose of hypoxia potentilla group significantly decreased and the LDH leakages increased. The levels of MCP-I,IL-1β, IFN-γ/and TNF-α were significantly increased,and the levels of VEGF, IL-6, IL-10 significantly decreased. Compared with the hypoxia group, the cell activities of each dose of hypoxia potentilla group significantly increased,and the LDH leakages decreased. The levels of cytokines reversed that the most obvious changes of MCP-1,1L-6,IFN-γ occurred in 1 g/L group, and the most obvious changes of 1L-1β, IL-10, TNF-α, VEGF occurred in 0.062 5 g/L group. Conclusion Potentilla anserine L. can antagonize the hypoxic damage effects of SH-SY5Y nerve cell induced by sodium dithionate in vitro.
