摘要
目的研究妊娠早期小鼠子宫内膜中IL-8RB在mRNA水平和蛋白质水平的表达情况。方法应用半定量RT-PCR、免疫组织化学显色和图像分析技术,观察IL-8RB在妊娠1d、4d、5d、6d小鼠子宫内膜中的表达情况。结果免疫组化染色结果显示在子宫内膜腔上皮,IL-8RB于妊娠4d表达最强,妊娠5d开始逐渐下降,妊娠6d降至妊娠1d水平;在子宫内膜腺上皮和基质,IL-8RB的表达水平随妊娠天数的增加而逐渐升高。RT-PCR结果显示IL-8RBmRNA表达规律与IL-8RB蛋白在子宫内膜腔上皮的表达相一致。结论 IL-8RB可能通过与其配体白细胞介素-8结合而参与小鼠胚泡着床过程。
Objective To study the expression of IL-8RB in mRNA and protein in mouse endometrium during early pregnancy. Methods The expressions of IL8RB in mouse endometrium on pregnant Day 1, Day 4, Day 5, and Day 6 were investigated by means of se- mi-quantitative RT-PCR, immunohistochemistry coloration method and image analysis techniques. Results Immunohistochemistry coloration results showed, in the luminal epithelium of endometrium, the expression of IL-SRB was the strongest on pregnant Day 4, then decreased gradually on Day 5, on Day 6 it reached the level of Day l.In the glandular epithelium and stroma of endometrium, the expression of IL-8RB increased gradually with the pregnant progress.RT-PCR results showed the expression of IL-8RB mRNA was consistent with the expression of IL-8RB protein in the luminal epithelium of endometrium.Conclusion IL-SRB might take part in the process of mouse blas- tocyst implantation by combining with its ligand interleukin-8.
