The 2012 Nobel Prize in Chemistry has been awarded to American scientists Robert J. Lefkowitz and Brian K. Kobilka groundbreaking discoveries that reveal the inner workings of an important family of such receptors: G-protein-coupled receptors (GPCR). The structure of GPCRs contains a seven-transmembrane domain, which pass through the cell membrane seven times. GPCRs are involved in a variety of physiological processes by sensing the ligand outside the cell and activating the downstream signal transduction pathway inside the cell. This review will summarize the structure and function of GPCRs and discuss their application in the treatment of human diseases and clinical medicine.
