摘要
通过动物实验探讨碱性成纤维细胞生长因子对角膜上皮细胞、基质细胞损伤的治疗作用。为受损角膜的愈合提供一条新的、有效的途径。方法 :将 2 4只兔全麻下切除角膜前板层 ,直径 8mm,约 1/ 3角膜厚度。将其随机分为 3组 ,分别滴生理盐水 ,0 .1μg/ ml及 1.0 μg/ ml碱性成纤维细胞因子溶液。术后定期用计算机图像处理系统测量角膜损伤后愈合面积 ,并用放射自显影方法观察角膜基质细胞生长及分布情况。结果 :术后 1~ 6日内 ,0 .1μg/ ml及 1.0 μg/ ml碱性成纤维细胞生长因子治疗组上皮愈合速率高于生理盐水 (对照 )组 ,且各治疗组未见显著的炎症反应及角膜新生血管浸润。术后第 3周末 ,实验组角膜 3H-胸腺嘧啶核苷标记阳性的基质细胞在创伤区增加较显著 ,并有集中趋势。结论 :0 .1~ 1.0μg/ ml的碱性成纤维细胞生长因子液滴眼可明显促进角膜损伤的修复 。
Objective:To investigate the effects of basic fibroblast growth factor(bFGF)on corneal wound of rabbit.Method:Anterior keratectomy of 8mm in diameter about 1/3 corneal thickness was performed on each eye of 24 rabbits.which were randomly divided into 3 groups.0 1,1 0μg/ml bFGF and normal saline(control)were applied respectively for each group.The healing area was determined by computer imagine analysis and proliferation and distribution of keratocytes were observed by the method of radioautography.Results:From 1~6 days after operation,the mean epithelial healing rates of 0 1,1 0μg/ml groups of bFGF were more than that the control group respectively.In the 3rd week,the keracytes of 3H TdR mark positive increased significantly in the wound area and had a tendency of collection.Conclusion:0 1~1 0μg/ml bFGF can promote corneal wound healing rate.It may be applied to serious corneal trauma clinically.
