人类CCK-45C/T遗传多态性及与抑郁症的相关性

摘要 目的调查胆囊收缩素-45C/T(CCK-45C/T)的遗传多态性,探讨其与抑郁症的遗传相关性。方法采集111例中国健康汉族人和130例抑郁症患者全血样本,利用TaqMan荧光标记探针杂交技术,使用Prism7900HT型荧光定量PCR仪,对CCK-45C/T进行基因频率调查,并比较分析健康人群和抑郁症患病人群CCK-45C/T等位基因的分布差异。结果 CCK-45C/T等位基因T的频率在健康群体中为0.369 4,在抑郁症患者群体中为0.496 2,组间比较存在显著性差异(P<0.05)。结论 CCK-45C/T等位基因T的分布在健康和抑郁症患者中差异显著,该基因可能与抑郁症易感相关。 Objective To investigate the polymorphism of cholecystokinin gene 45C/T locus (CCK- 45C/T) in promoter region and potential association between the polymorphism and depression. Methods We collected the whole blood samples of 111 Chinese healthy Han individuals and 130 individuals with depression, using TaqMan probe labeled with fluorescent dye hybridization technique by ABI Prism 7900HT Sequence Detector , to investigate the gene frequency of CCK-45C/T. Results The T allele frequency of CCK- 45C/T was 0. 496 2 in depressive individuals and 0. 369 4 in healthy ones. A significant difference between the two groups was observed ( P 〈 0. 05 ). Conclusion There was a significant difference in the T allele frequency of CCK-45C/T between the healthy and depression individuals. It was indicated that the T allele of CCK-45C/T may be a potential susceptibility allele in depression.
