摘要
本文报告了从抗日本血吸虫单抗NP11- 4杂交瘤细胞株的基因组DNA中 ,用PCR技术扩增出抗体轻链可变区基因 ,并克隆至pUC19质粒中进行测序分析。研究表明 :该基因长 336bp ,编码 112个氨基酸 ,经结构分析及与抗体库中一般序列的比较 ,证实该序列为轻链可变区基因 ,属κ轻链SubgroupII亚类 ,由种系基因中的V与Jκ 2基因重排而成。
The V L gene of a McAb against the membrane protein of Schistosoma japonicum was amplified from the genomic DNA of the hybridoma NP11-4 by PCR The PCR products were cloned into pUC19 Vector Several recombinants that had been inserted with the target fragment were sequenced by Sangers method The result showed that the V L was 336bp long, encoding 112 amino acids It belongs to the mouse Vκ subgroup II and rearrangs from V gene and Jκ 2 gene
