孕期营养不良对宫内发育迟缓大鼠心肌重构的影响 被引量:3

Effects of gestational malnutrition on cardiac remodeling of intrauterine growth retardation rats

摘要 目的观察孕期营养不良对宫内发育迟缓(IUGR)大鼠心肌重构的影响。方法 6只SD雌性大鼠随机分成正常对照组和IUGR组,每组3只。采用妊娠全期饥饿法建立IUGR动物模型,分别有16只正常仔鼠和16只IUGR仔鼠纳入实验对象。仔鼠3周时断乳,16周时测定各组仔鼠左心室重量指数(LVMI),免疫组化法检测心肌I型和III型胶原表达。结果与正常仔鼠相比,IUGR仔鼠出生时平均体质量和身长明显落后(P<0.01)。给予正常饮食后,IUGR仔鼠出生后呈现追赶生长现象,并于第15周赶上正常对照组体质量水平。IUGR组仔鼠的LVMI较正常对照组明显增大(P<0.05),其心肌I型和III型胶原表达量均显著增高(P<0.01)。结论孕期营养不良可导致IUGR仔鼠心肌发生重构,出生后正常饮食亦未能干预心肌重构的发生和发展。 Objective To investigate the influence of gestational malnutrition on myocardial remodeling of intrauterine growth retardation (IUGR) rats. Methods Six SD female rats were randomly divided into control group and IUGR group. The IUGR animal model was established by whole term maternal nutrition restriction. There were 16 normal and 16 IUGR newborn rats in the experiment. The newborn rats were weaned 3 weeks after birth, and their left ventricular mass index (LVMI) was measured, and expression of collagen I and HI was assessed by immunohistochemistry at 16 weeks of age. Results Compared with control group, average body weight and length decreased in IUGR group (P〈0.01). After fed normal diet, IUGR rats showed a catch-up growth phenomenon and their body weight came up to that of control group at 15 weeks of age. LVMI and expression of myocardial collagen I and III were significantly higher in IUGR group than those in control (P〈0.01 or 0.05). Conclusion Gestational malnutrition leads to myocardial remodeling of IUGR rats. Postnatal normal nutrition is unlikely to prevent the development and progression of cardiac remodeling in IUGR rats.
