摘要
目的研究吗啡与5-氟尿嘧啶(5-Fu)联合应用对MCF-7乳腺癌细胞Bcl-2、Bax表达的影响。方法药物作用48 h后通过免疫细胞化学技术检测Bcl-2、Bax表达情况。结果 Bcl-2表达情况:未处理组最强。单独应用吗啡处理时,浓度为250、1 250μmol/L引起Bcl-2表达减弱(P<0.05,P<0.01),三种浓度的吗啡与5-Fu联合应用均具有协同作用(P<0.05)。Bax表达情况:未处理组最弱。单独应用吗啡处理时,250、1 250μmol/L浓度的吗啡引起Bax表达增强(P<0.05,P<0.01),三种浓度的吗啡与5-Fu联合应用,均具有协同作用(P<0.05)。结论一定浓度的吗啡能与5-Fu发挥协同作用,强化5-Fu诱导的Bcl-2表达下调、Bax表达上调,且与吗啡的浓度有一定的剂量依赖关系。
Objective To study on the effects of different concentrations of morphine on Bcl-2 and Bax expressions in the process of MCF-7 cellular apoptosis induced by 5-Fluorouracil. Methods After treated for 48 h, the expressions of Bcl-2 and Bax were detected by immunocytochemistry. Results The expression of Bcl-2 of untreated group was the strongest. Using 500 p.mol/L 5-FU only, the expression of Bcl-2 was decreased (P 〈 0. 01 ). Using morphine only, 250 and 1 250 μmol/L of morphine leaded to decreasing the expression of Bcl-2 (P 〈 0. 05 ,P 〈 0. 01 ). Using morphine combined with 5-FU, the synergisms of morphine with 5-FU were found in all three groups (P 〈 0. 05 ). The expression of Bax in untreated group was the weakest. Using 500 μmol/L 5-FU only, the expression of Bax was increased(P 〈 0. 01 ). Using morphine only, 250 and 1 250 μmol/L of morphine leaded to increasing the expression of Bax (P 〈0. 05 ,P 〈0. 01 ). Using morphine combined with 5-FU, the synergisms were found in all three groups (P 〈 0. 05). Conclusions Morphine in certain concentration could enhance significantly the down-regulating effect on Bcl-2 expression and up-regulating effect on Bax expression that are induced in MCF-7 breast cancer cells, in certain dose dependent manner.
