摘要
研究了利福平(rif)对大肠杆菌F′质粒的消除效应.实验结果表明,消除质粒以rif的浓度50—100μg/mL为宜.通过选择培养,在LB和LB+cm平板上,就能选出F^-菌株,因F^+抗cm,E^-不抗cm.选得的F^-菌株是:E.Coli XAC F^-/△(lac pro)nal.
This paper reports an efficient method of F' plasmids curing with rifampin. The replication of plasmids can be selectively inhibited by rifampin. Crowth of strains carrying F or F' factors in rifampin therefore results in loss of the sex factor. The result shows that the concentrations of rifampin of 50-100ng/mL are widely used for this purpose. In this method, only by using two selectivity medium can the strains of F+ or F- be selected. Thus, the selection is for E.coli XAC F-/A (lar pro) nal.
