摘要
目的 :探讨自身抗原La的抗原优势表位 ,为疾病机制的研究提供依据。方法 :根据计算机软件进行的蛋白质序列结构分析 ,采用PCR法克隆自身抗原La多肽片段的cDNA ,定向插入表达载体PGEX 2T ,并且导入大肠杆菌中表达重组融合蛋白 ,用GST亲合层析柱进行纯化 ,经免疫印迹法与患者阳性血清进行反应。结果 :SSB/La的抗原优势表位主要存在于1~ 2 8,80~ 10 7,10 8~ 188,2 83~ 338位。不同病人在不同病程的La优势表位有所不同。结论 :抗原驱动机制在自身免疫疾病的发病中起重要作用 。
Objective: The interactions between autoantigen and autoantibody play an important role in the pathogenesis of autoimmune diseases. In view of analysis of the epitopes of autoantigen SSB/La to understand the molecular mechanism of autoimmune diseases. Methods:Based on the sequence analysis of autoantigen SSB/La, the seven fractions of SSB/La GSTantigen fusion proteins induced by IPTG in E.coli.JM109 were obtained.The epitopes of SSB/La were analyzed by IBT and competitive inhibition reaction. Results:The epitopes of SSB/La were existed on aa 1~28,80~107,108~188,283~338.Conclusion:The antigendriving mechanism play an important role in the pathogenesis of autoimmune diseases.
