氯胺酮和γ-羟基丁酸钠对家兔支气管平滑肌腺苷酸环化酶和磷酸二酯酶活力的影响 被引量:2

摘要 目的 :研究氯胺酮和γ 羟基丁酸钠对支气管平滑肌腺苷酸环化酶 (AC)和磷酸二酯酶 (PDE)活力的影响。方法 :离体实验 ,分为对照组、氯胺酮组和γ 羟基丁酸钠组 ,每组 8只家兔。氯胺酮和γ 羟基丁酸钠的实验浓度分别为2 3× 10 -4 M和 3 0× 10 -3 M。结果 :氯胺酮和γ 羟基丁酸钠对AC活力无明显影响 ,但均可显著抑制PDE的活力。两组PDE活力分别为 2 42± 0 6 3U和 3 6 0± 0 87U ,与对照组 4 5 8± 0 42U相比 ,分别P <0 0 1和P <0 0 5。结论 :氯胺酮和γ 羟基丁酸钠均可减少支气管平滑肌中cAMP的降解 ,这可能是氯胺酮和γ 羟基丁酸钠在麻醉中使支气管平滑肌张力下降的原因之一。 Objective:To explore the effects of ketamine and γ hydroxybutyrate sodium(γ OH) on adenyl cyclase(AC) and phosphodiesterase(PDE)of the rabbit bronchial smooth muscle in vitro.Methods:The rabbit’s bronchial smooth muscle samples were divided into control group,ketamine group and γ OH group with 8 samples each.The reactive concentration of ketamine was 2 3×10 -4 M or γ OH was 3 0×10 -4 M.Results:The activity of AC was not affected by ketamine or γ OH, but the activity of PDE could be inhibited by ketamine or γ OH.The activity of PDE was 2 42±0 63U in ketamine group and 3 60±0 87U in γ OH group.Compared with control group 4 58±0 42U,the activity of PDE was obvious lower in ketamine group ( P< 0 01) or in γ OH group ( P< 0 05).Conclusion:Both ketamine and γ OH can slow down the degradation of cAMP.This may be one of the reasons for lowering the tone of the bronchial smooth muscle during anesthesia.
