摘要
目的 建立反相高效液相色谱法测定人血浆中氢溴酸右美沙芬的浓度。方法 采用Nova PakC18色谱柱 ,以乙腈 水 36 %乙酸 (5 0∶49∶1) ,用三乙胺调 pH至 4.3,流速 :1ml·min-1,荧光检测 ,激发波长 2 80nm ,发射波长 310nm。结果 氢溴酸右美沙芬约 1.95min达高峰浓度 ,cmax为 (6 .2 0± 1.32 )ng·ml-1。结论 本法简便、灵敏、专属性好 。
OBJECTIVE To develop a RP HPLC assay for the determination of dextromethorphan hydrobromide (DM) in healthy volunteers.METHODS The analytical colunm was packed with Nova Pak C 18 .The mobile phase consisted of a mixture of acetonitrile water acetic acid (50∶49∶1).Triethylamine was used to adjust pH to 4.3.Flow rate was 1 ml·min -1 .Fluor escence detector was used with Ex at 280 nm and Em at 310 nm.RESULTS: T max ws about 1.95 min. c max was (6.20±1.32) ng·ml -1 .CONCLUSION:The assay here was sensitive,simple and accurate,can be used to study pharmacokinetics of DM.
