摘要
目的:考察pH、温度、光照等因素对胸腺肽α1稳定性的影响。方法:采用HPLC法分析胸腺肽α1在不同条件下的含量变化,将浓度变化与时间作线性回归,求得药物在不同pH条件下的降解曲线方程,利用降解速率常数反映出胸腺肽α1在恒温条件下的最适pH。结果:酸、碱、光照及高温均能促进胸腺肽α1的降解,药物的最稳定pH在5~6之间,且在醋酸盐缓冲液中较稳定。结论:温度、pH、光照等因素对胸腺肽α1的稳定性有显著影响。
Objective:To investigate the influence of pH, temperature and light on stability of thymosin α1. Meth- ods:An adjusted HPLC method was employed to determine concentration changes of thymosin α1 with these experi- mental factors mentioned above. The degradation calculations of thymosin α1 under different pH conditions were ob- tained by linear regression analysis between concentration and time, whose slopes indicated the sensitivity of the drug stability to these influencing factors. Result:Acid, alkali, light and high temperature could promote the degra- dation of thymosin α1. Thymosin α1 was stable in the acetate buffer, and achieved the highest stability when pH was between 5 and 6. Conclusion:Temperature, pH, light and other factors have a significant effect on the stability of thymosin α1
