摘要
目的探讨livinβ基因阻断对胶质瘤细胞增殖和凋亡能力的影响。方法脂质体法转染重组质粒于人星形胶质瘤细胞(U251),MTT法检测细胞增殖,流式细胞仪检测细胞凋亡值,RT-PCR、Western blot分别检测细胞中caspase-3基因及蛋白的表达。结果 MTT法检测转染组细胞增殖明显低于对照组和空载体组,差异有统计学意义(P<0.05);PI单染流式细胞术检测转染组细胞凋亡值明显低于对照组和空载体组,差异有统计学意义(P<0.05);转染组caspase-3基因及蛋白表达均明显高于对照组和空载体组,差异有统计学意义(P<0.05)。结论阻断人星形胶质瘤细胞(U251)中livinβ基因表达,caspase-3基因及蛋白表达均明显增加,激活细胞凋亡蛋白酶级联反应,导致凋亡增加,细胞增殖减少,进一步证实了livinβ基因在胶质瘤的发生、发展中起着重要作用。
Objective To investigate whether silencing livinβ gene by siRNA leads to apoptosis,arrest of cell cycle and inhibition of proliferation.Methods The recombinant plasmid was transfected into U251 glioma cells by liposome mediated transfection.Then PI flow cytometric analysis was performed for cell cycle and apoptosis,MTT assay for cell proliferation,RT-PCR and Western blotting for caspase-3 expression.Results Silencing livinβ gene by siRNA could inhibit cell proliferation remarkably(P0.05),could arrest cell cycle significantly and induce cell apoptosis(P0.05),could upregulate caspase-3 obviously(P0.05).Conclusion Silencing livinβ gene in glioma cells could activate caspase-3 and increased poly(ADP-ribose) polymerase cleavage,therefore induce apoptosis and inhibit proliferation.So the livinβ may play key role in pathogenesis of glioma.
