摘要
目的:观察临床有效的益气活血解毒代表方药——化瘀丸联合化疗对肿瘤生长及血栓形成相关因子的影响。方法:建立小鼠Lewis肺癌动物模型,实验分为模型组、环磷酰胺(CTX)组、CTX+低分子肝素组、CTX+化瘀丸高剂量组、CTX+化瘀丸低剂量组、空白对照组。定期观察各组小鼠体质量及肿瘤生长情况,接种后第13、20天处死小鼠,检测瘤重、肺转移数,ELISA法检测小鼠血清肿瘤坏死因子-α(TNF-α)、P-选择素(P-selectin)、内皮素-1(ET-1)水平。结果:第20天,CTX+化瘀丸高剂量组小鼠体重显著高于模型组及CTX组(P<0.01);各化疗组均有明显的肿瘤抑制作用和肺转移抑制作用,但组间无差异;CTX+化瘀丸高剂量组小鼠血清P-selectin水平显著低于CTX组(P<0.01)。结论:高剂量化瘀丸联合CTX化疗能够显著改善Lewis肺癌小鼠体质量下降的趋势,并能显著下调化疗引起的P-selectin水平异常升高。
Objective： To observe the effect of Huayu Wan combination chemotherapy on tumor growth and expression of thrombosis formation correlation factors in Lewis lung cancer-bearing mice.Methods： Lewis lung cancer model in mice was established and divided into 5 groups： model group,CTX group,Low-Molecular-Weight Heparins＋CTX group,Huayu Wan high dose＋CTX group,Huayu Wan low dose＋CTX group.There was also a group of normal mice for negative control.The tumor inhibiting rate,the inhibiting rate of lung metastasis and change of weight were observed regularly in 13 and 20 days after inoculate and the levels of TNF-α,P-Selectin,ET-1 in peripheral blood serum were tested by ELISA.Results： The increase of body weight was obvious in Huayu Wan high dose＋CTX group（P0.01）（20d）.The tumor inhibition rate and lung metastasis inhibition rate were significant in all chemotherapy groups.The level of P-selectin in peripheral blood serum of Huayu Wan high dose＋CTX group was lower than that in CTX group（P0.01）（20d）.Conclusion： High dose of Huayu Wan combination chemotherapy can be an effective measure for increasing the body weight and reducing the expression of P-selectin in Lewis lung cancer-bearing mice.
