摘要 Asp .NigerNo .3单宁酶通过超滤、DEAE -cellulose离子交换层析 ,SephadexG- 1 50柱层析以及Sepharos- 6B柱层析四步纯化后 ,经过SDS 聚丙烯酰胺凝胶电泳鉴定 ,显现一条蛋白质带 ,酶的总活力回收率达到1 1 % ,纯化倍数为 1 1 3 5 ;对酶的贮藏稳定性、温度稳定性、pH稳定性、酶促反应的最适pH值以及最适反应温度进行了研究。 Tannase was purified about 113.5-fold from the mycelia of Aspergillus Niger No.3 by a process involving ultrafiltration.DEAE-cellulose column chromatography,sephadex G-150 gel filtration and Sepharose-6B gel filtration.11% of the tannase was recovered.The purified tannase appeared only a band on the diagram of SDS-polyacrylamide gel electrophoresis.Some properties of tannase were studied.
