摘要 利用实时荧光定量技术,采用60Coγ射线照射人肝细胞株,照射剂量为0、0.1、0.2、0.5、1.0、2.0、4.0 Gy,观察比较照后不同时间胰岛素生长因子1受体(IGF-1R)的表达水平的改变,探讨IGF-1R基因的剂量-效应和时间-效应关系。结果表明:①人肝细胞受照后,IGF-1R基因的不同剂量组在3个时间点的相对表达量均小于1,均为下调表达;②照后6小时,IGF-1R基因下调表达水平先下降后上升,照后12小时和24小时,随着照射剂量的增加,IGF-1R表达水平均呈现先升高后降低的趋势,其中,照后12小时在1 Gy时表达水平最高,照后24小时在0.5 Gy时最高;③剂量-效应和时间-效应关系的相关性均不显著。 This study is to observe and compare the changes in IGF-1R gene expression in Hepatocytes irradiated by 60Co gamma rays with doses of 0, 0.1, 0.2, 0.5, 1.0, 2.0 and 4.0 Gy at different time-points by using the technology of real-time fluorescent quantitative PCR, so as to explore IGF-1R gene doses-effect relationships and time-effect relations. The result shows that: (1) after the irradiation, relative quantitative ratio of IGF-1R in different doses at three time points are less than 1, so they are down-regulation comprehensively, (2) after irradiation 6 hours, there was no significant dose-effect relationship. In the 12 h and the 24 h groups, with the increment of irradiation doses, IGF-1R level was enhanced gradually and then decreased in all time groups. After irradiation of 1 Gy, the expression levels of IGF-1R peaked at 12 h. After irradiation of 0.5 Gy, the expression of these genes peaked at 2A h.
