摘要
通过 p H吸附法 ,利用乳酸链球菌亚种 N8(L actococcus lactis subsp. lactis N8)和乳酸链球菌亚种ATCC1145 4,研究了 nisin Z分子在不同 p H环境下的吸附作用及其分离纯化效率 ,并经麦黄酮 SDS- PAGE和 RP- HPL C纯度鉴定 ,证实用此技术分离 nisin Z具有高效高纯度、操作简便等良好特征。nisin在 p H6 .5吸附率达 10 0 % ,p H3以下完全释放。该技术可望开辟对 nisin等微生物分泌性蛋白的分离纯化新途径。
By using pH adsorption and making use of Lactococcus lactis subspecies lactis N8 and ATCC 11454, the adsorption and efficiency of isolation and purification of nisin Z molecule at different pH condition were studied. Identification of tricine sodium dodecyle sulfate polyacrylamide gel electrophoresis and reverse phase high pressure liquid chromatography showed that the technique of isolation and purification by pH adsorption was characterized by high efficency, high purity of molecules and simple in manipulation. Rate of nisin adsorption to cells reached 100 % at pH 6.5 and the nisin molecules were completely released at pH 3. The technique is hopeful to be used as a new way for isolating and purifying the released small bacteriocins like nisin.
