摘要
目的 探讨 HPV16 E6反义寡核苷酸 (AE6 )对宫颈癌细胞 Si Ha细胞凋亡的诱导作用 .方法 应用细胞生长抑制实验 (MTT)、透射电子显微镜 (TEM)、DNA电泳及流式细胞术 (FCM)研究 AE6对 Si Ha细胞生长抑制、超微结构改变、核酸及细胞增殖周期的影响 .结果 AE6 1~ 40μmol· L- 1对 Si Ha细胞抑制率为 9.9%~ 6 0 .3% ,相互间有显著性差异(P<0 .0 5 ) .AE6 2 0 μmol· L- 1作用 72 h,Si Ha出现凋亡形态学改变 ,DNA电泳呈梯状带型 .AE6 5 ,10 ,2 0和 40μmol· L- 1 作用 72 h后 ,Si Ha细胞凋亡率分别为 1.9% ,7.9% ,8.1%和 19.7% .结论 HPV16 E6基因与 HPV16阳性宫颈癌细胞的增殖和细胞凋亡密切相关 ,AE6可通过诱导 Si Ha细胞凋亡而抑制其生长 。
AIM To investigate apoptosis in SiHa induced by antisense oligonucleotide of human papillamavirus type 16 E6 gene. METHODS MTT, transmission electron microscope (TEM), DNA gel electrophoresin and flow cytometry (FCM) were performed to examine the proliferation inhibi tion, morphological appearance as well as DNA and cell cycle treated with AE6 in SiHa. RESULTS The inhibition rates of AE6 1 μmol·L -1 to 40 μmol·L -1 were 9.9%~60.3%. After the treatment with AE6 20 μmol·L -1 for 72 h, apoptotic appearance and DNA ladder were observed. When AE6 5, 10, 20 and 40 μmol·L -1 were given for 72 h, the percentages of apoptotic cells were 1.9%, 7.9%, 8.1% and 19.7% respectively. CONCLUSION HPV16 E6 gene may play an important role in the carcinogenesis and apoptosis of HPV16 positive cervical carcinoma cell SiHa. AE6 can inhibit the proliferation of SiHa by the way of apoptosis.
