摘要
目的 研究 p73基因在结肠癌中的改变及其意义 .方法 采用 PCR- SSCP方法检测 40例结肠癌及其癌旁非癌组织中 p73基因突变 ,采用定量 RT- PCR方法检测 p73基因的表达 .结果 1PCR- SSCP检测出 7例存在 p73等位基因缺失 ,未见突变 ;2定量 RT- PCR检测出 33例结肠癌呈高表达 ,7例呈中等表达 ;非癌组织中 2 6例呈低水平表达 ,14例呈中等水平表达 ;p73基因在结肠癌与癌旁组织中的表达存在显著性差异 (P<0 .0 1) .结论 p73基因以等位基因缺失。
AIM To investigate the alteration of p73 gene in colon cancer and paracancerous tissues and its biological significance. METHODS 40 specimens of colon cancers and matched paracancerous tissues were screened for mutation by PCR SSCP and for expressional levels of p73 gene by quantitative RT PCR. RESULTS ① Seven cases of colon cancers were screened to have allele loss by PCR SSCP and no mutation was observed. ② By RT PCR, 33 specimens of colon cancers were screened to be overexpressed, 7 specimens of colon cancers were medially expressed, 26 specimens of paracancerous tissues were lowly expressed and 14 cases were medially expressed. There was significant difference between colon cancers and paracancerous tissues ( P <0.01). CONCLUSION p73 gene may take part in the development of colon cancer.
