摘要
目的 研究 MN92 0 2 (6 -二甲基 - 4- (3-硝基苯基 ) - 1,4-二氢 - 3,5 -吡啶二羧酸 - 3-甲酯 - 5 -正戊酯 )对高脂血症和动脉粥样硬化的治疗效果及其机制 .方法 40只 SD大鼠随机分为正常组 ,高脂组 ,MN92 0 2 1,3和 5 mg· kg- 1 组 .每日记录饮食 ,体质量 ;每月测量血压 .3mo后测血浆胆固醇 (CT) ,甘油三脂 (TG) ,低密度脂蛋白 (L DL- c) ,高密度脂蛋白 (HDL- c)血清丙二醛 (MDA)和超氧化物歧化酶 (SOD) ,并用光镜和电镜观察主动脉的组织学变化 .结果 高脂组 CT,L DL - c,MDA明显升高 .MN92 0 2 3mg· kg- 1 和 MN92 0 2 5 mg·kg- 1 组 CT值从 (10 .7± 1.6 ) mmol· L- 1 分别降到 (6 .9±2 .4) m mol· L- 1 和 (5 .2± 2 .1) mmol· L- 1 ,L DL - c从 (7.6±2 .0 ) mmol·L- 1分别降到 (3.1± 1.0 ) mmol· L- 1和 (2 .6±1.3) mmol· L- 1 .MDA从 (2 .9± 1.0 ) mmol· L- 1分别降到(1.4± 0 .9) mmol· L- 1 和 (1.2± 0 .7) mmol· L- 1 .组织形态学显示 ,高脂组动物主动脉壁不完整 ,内皮细胞出现空泡 ,平滑肌细胞增殖 ,并见脂滴 .MN92 0 2 5 mg· kg- 1组的上述变化较对照组和其他给药组明显减轻 .结论 MN92 0 2抗高脂血症和阻止动脉粥样硬化发展的机制 ,可能与降低血浆CT和 L DL -
AIM To investigate the effects of MN9202 on experimental hypercholesterolemia and atherosclerosis in rats fed with high lipid diet. METHODS Forty Sprague Dawley rats were randomly divided into five groups: A normal control group with rats fed with standard chow diet, and four other groups fed for three months with a high fat diet plus saline, vehicle and MN9202 at the doses of 1, 3 or 5 mg·kg -1 /day. Dietary consumption and body weight were recorded everyday, and blood pressure was measured every month. The levels of plasma cholesterol (CT), triglyceride (TG), high density lipoprotein cholesterol (HDL c), low density lipoprotein cholesterol (LDL c), malondialdehyde (MDA) and superoxide dismutase (SOD) were determined. Morphological changes of aorta were observed under light and electronic microscopes at the end of the experiment. RESULTS There was a significant increase of CT, LDL c and MDA in high lipid diet fed animals. MN9202 at the doses of 3 and 5 mg·kg -1 could decrease CT from (10.7±1.6) mmol·L -1 to (6.9±2.4) and (5.2±2.1) mmol·L -1 ( P <0.05, P < 0.01 ), LDL c from (7.64±2.03) to (3.08±1.01) and (2.63±1.25) mmol·L -1 ( P <0.05, P <0.01), respectively and MDA level from (2.90±1.0) mmol·L -1 to (1.4±0.9) and (1.2±0.7) mmol·L -1 ( P <0.05, P <0.01) respectively. Morphologically, there were impairments in the endothelial cells of the aorta in the lipid diet fed group, which became less notable with MN9202 at high doses. CONCLUSION MN9202 has anti hypercholesterolemia effect and can inhibit the development of atherosclerosis, which may be due to its suppressive effect on CT, LDL c and lipid peroxidation.
