摘要
目的 建立大鼠 C6脑胶质瘤模型 ,行一般观察和MRI检查以确定颅内肿瘤生长情况 ,寻找简易判定颅内肿瘤大小的可靠指标 .方法 体外培养大鼠 C6胶质瘤细胞 ,应用立体定向法将含 10 g· L- 1 琼脂糖和 1× 10 6 C6细胞悬液 10μL注入大鼠右脑尾状核 .接种后行一般观察和 MRI检查 ,对5组接种大鼠分别在第 10 ,15 ,2 0 ,2 5日和自然死亡前做主动脉多聚甲醛灌注固定 ,对脑组织和肿瘤标本进行切片观察和HE染色 .结果 5 0只接种大鼠均有脑内肿瘤生长 (10 0 % ) ,远处和颅外转移率为 0~ 4% .结论 成功建立大鼠 C6脑胶质瘤模型 ,接种后脑内肿瘤呈球状生长 ,很少出现颅外转移 .大鼠的生存时间可作为判定颅内肿瘤生长大小的指标 .
AIM To establish a rat C6 brain tumor model, observe the tumor growth by general observation and MRI scan and seek for a simple and reliable index to judge its size. METHODS Rat C6 gliomas cells were cultured in vitro . 10 μL C6 cell suspension containing 10 g·L -1 agarose and 1×10 6 cells was stereotaxically injected into the right caudatenucleus of the rat brain. Implantation was followed by general observation and MRI scan. In the 5 groups with implantation, trans aorta paraformaldehyde perfusion was conducted at 10, 15, 20 and 25 days following the implantation and before the natural death. Brains, lungs, spinal cords and tumors were sectioned and inspected. The tumor containing samples were prepared histologically by hematoxylin and eosin (HE) stains. RESULTS Intracerebral growth occurred in the 50 implanted rats (100%), with a distant metastasis of 0 to 4%. CONCLUSION The rat C6 brain tumor model is successfully established. After implantation, globular intracrerbral growth occurs and extracranial growth extension is scarce. The implanted rat survival duration is correlated with the size of the tumor and can be used as an index of its size.
