摘要
目的:观察芪黄明目胶囊对糖尿病小鼠视网膜氧化应激的作用并探讨其机制。方法:KK/Upj-Ay小鼠随机分为模型组、芪黄明目高、中、低剂量组,另设C57BL/6小鼠对照组。灌胃给药3个月,观察一般情况,测定空腹血糖(FBG);检测血液中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)、过氧化氢酶(CAT)含量;光镜及电镜观察视网膜形态学变化;Western blot法测定视网膜p38MAPK、p-p38MAPK、JNK、p-JNK、ERK、p-ERK蛋白的表达。结果:芪黄明目能不同程度改善模型小鼠症状,降低FBG;减轻视网膜病理损伤;不同程度的升高SOD、GSH-Px、CAT活性,降低MDA含量;与模型组比较,芪黄明目组p38MAPK蛋白磷酸化水平明显下降(P<0.01);JNK和ERK蛋白磷酸化水平比较无显著差异;p38MAPK、JNK、ERK蛋白在各组表达无统计意义。结论:芪黄明目可通过抑制p38MAPK磷酸化水平而实现其抗氧化应激、保护视网膜的作用。
Objective: To investigate the effect of qihuangmingmu capsule(QHMM) on oxidative stress of retina in diabetic mice and its mechanisms.Methods: KK/Upj-Ay mice were randomly divided into model,QHMM(low,middle,high).C57BL/6 mice were selected as control group.Mice were given intragastrically for 3 months.The general appearance,fasting blood-glucose(FBG) were observed.Activity of SOD,GSH-Px,CAT and content of MDA in blood were detected.Pathological changes of retina were observed by light microscope and electron microscope.The expression of p38MAPK、p-p38MAPK、JNK、p-JNK、ERK、p-ERK in retina were examined by Western blot.Results: QHMM could ameliorate the symptom of DM mice.FBG were decreased.Pathological lesions of retina were alleviated in QHMM group.Activity of SOD,GSH-Px,CAT were increased,while content of MDA were decreased.The phosphorylation of p38MAPK were significantly decreased(P0.01).The phosphorylation of JNK and ERK were not significant deviation.Conclusion: QHMM has protective effects on diabetic retinopathy of mice via inhibiting oxidative stress and suppressing the phosphorylation of p38MAPK.
