头穴丛刺法对脑缺血再灌注大鼠脑组织AQP_4表达的影响 被引量:7

The Effect of Cluster Acupuncture on Scalp Points on AQP_4 Expression in Cerebral Ischemica Reperfusion Rats

摘要 目的:研究头穴丛刺法对脑缺血再灌注大鼠的脑保护作用机制。方法:采用改良的线栓法制备大鼠大脑中动脉缺血再灌注模型。将Wistar雄性大鼠,随机分成3组:假手术组、模型组、头穴丛刺组。各组随机分成6 h、1 d、2 d、3 d共5个时间点,采用免疫组织化学技术检测大鼠脑缺血再灌注后AQP4蛋白表达变化及头穴丛刺法治疗对它们的影响。结果:免疫组化结果显示与模型组比较,头穴丛刺组损伤侧脑组织AQP4蛋白表达水平明显升高(P<0.05或P<0.01)。结论:头穴丛刺法能够调节AQP4的表达,减轻由于再灌注损伤导致的血脑屏障通透性增强,从而减轻脑水肿,在脑缺血再灌注早期起到挽救和保护受损神经元、改善神经功能的治疗作用。 Objective:To explore cerebral protection mechanism of cluster acupuncture on scalp points(CASP) in cerebral ischemia reperfusion(CIR) rats.Methods:Focal cerebral ischemia was induced by intraluminal middle cerebral artery occlusion(MCAO) method using a monofilament thread in rats.Male Wistar rats were divided randomly into three groups: sham group、model group、and CASP group;each group was separated into 6 h,l d,2 d,3 d,5 d after reperfusion as subgroup for investigation.The expression of AQP4 protein in CIR rats were measured by immunohistochemical method and the influence of the expression by CASP was observed.Results:Immunohistochemistry results indicated that compared with model group,the expression of AQP4 has been significantly increased in CASP group(P0.05,P0.01).Conclusion:CASP method can regulate AQP4 expression to reduce BBB permeability to prevent cerebral edema,through reducing cerebral edema,CASP method can save and protect damaged neurons,improve nervous function at early stage of CIR.
