摘要
目的分析IRM-2小鼠全长cDNA文库的cDNA序列。方法根据IRM-2小鼠的21条EST,从IRM-2小鼠全长cDNA文库测定基于该21条EST序列PCR扩增的全长cDNA序列,通过与GenBank基因库同源序列信息比对,分析全长cDNA序列和性质。结果根据21对引物PCR产物的拼接,获得5条全长cDNA序列,与小鼠已知基因不同源,而且得到了可能编码蛋白质的氨基酸序列和蛋白质的性质。结论 5条全长cDNA序列提示IRM-2小鼠体内可能存在辐射抗性相关基因,为进一步分离和鉴定辐射抗性相关基因奠定了基础。
Objective To analyse cDNA sequences from full-length cDNA library of IRM-2 mice.Methods Full-length cDNA products were amplified by PCR from cDNA library of IRM-2 mice according to twenty-one pieces of EST.Full-length cDNA sequences and properties were analyzed by comparing with GenBank database.Results Five pieces of full-length cDNA were obtained according to PCR products amplified by twenty-one pairs of primers,which were not the same as that of known mice genes.Amino acid sequences and properties of proteins encoded by five pieces of full-length cDNA were acquired.Conclusion Five pieces of full-length cDNA imply that radioresistance correlative genes may be existed in IRM-2 mice,laying a foundation for isolating and identifying radioresistance correlative genes in further study.
