摘要
目的:探讨mina53基因在胃癌组织中的表达及其与临床分期的相关性。方法:分别应用RT-PCR和免疫组化技术,检测71例胃癌患者组织标本以及20例癌旁组织中mina53基因和蛋白的表达水平,并分析其与胃癌临床分期的相关性。结果:(1)RT-PCR结果显示20例癌旁组织中仅2例mina53基因呈弱阳性表达,阳性率为10.00%;71例胃癌组织标本中有43例mina53基因表达阳性,阳性率为60.56%(P<0.05);其中I、Ⅱ、Ⅲ、Ⅳ期胃癌组织标本中的阳性比例分别为11/23、15/29、12/14及5/5(P<0.05)。(2)免疫组化结果显示20例癌旁组织中仅3例mina53蛋白呈弱阳性表达,阳性率为15.00%;71例胃癌组织标本中有35例mina53蛋白表达阳性,阳性率为49.30%(P<0.05);其中I、Ⅱ、Ⅲ、Ⅳ期胃癌组织标本中的阳性比例分别为7/23、13/29、11/14及4/5(P<0.05)。结论:与癌旁组织相比,mina53在胃癌中的基因和蛋白水平存在高表达,并且随肿瘤分期的进展而表达量增高,检测mina53表达水平有助于胃癌早期诊断和预测肿瘤的侵袭程度。
Objective: To investigate the correlation of mina53 gene expression in gastric cancer tissues with the clinical phrase. Method: The RT-PCR and immunohistochemistry methods were applied to detect the gene and protein expression of mina53 in gastric cancer tissue of 71 patients and 20 samples of adjacent mucosa, and to analyze its correlation with the gastric cancer stage. Results: (1) RT-PCR results showed 2 out of 20 cases of adjacent noncancerous tissue had weakly positive expression ofmina53 gene, with a positive rate of 10.00%, and that 43 out of 71 cases of gastric cancer tissue specimens had positive expression of mina53 gene, with a positive rate of 60.56%. Among the gastric cancer tissues, 11 out of 23 cases of phase I, 15 out of 29 cases of phase II, 12 out of 14 cases of phase III, and 5 cases of phase IV showed positive expression of mina53 gene(P〈0.05 ). (2) Immtmohistochemistry results showed that among the 20 cases of adjacent noncancerous tissue, only three cases had weakly positive protein expression of mina53, with a positive rate of 15.00%; and among the 71 cases of gastric cancer tissue specimens, 35 cases had positive protein expression of mina53, with a positive rate of 49.30%. Among the 71 cases, 7 out of 23 cases of phase I, 13 out of 29 cases of phase II, 11 out of 14 cases of phase III, and 4 out of 5 cases of phase IV showed positive mina53 protein expression (P〈0.05). Conclusion: Compared with the adjacent tissues, mina53 gene and protein were highly expressed in gastric cancer, and their expression levels increased with tumor stage progress. Detection of mina53 expression levels is conducive to early diagnosis of gastric cancer and prediction of tumor invasion.
