摘要
本实验用聚合酶链反应与随机引物标记法制备地高辛素标记的肾综合征出血热病毒核酸M、S片段cDNA探针。DNA-RNA斑点杂交结果表明，该探针具有肾综合征出血热病毒的特异性，可检出10pgHFRSV-RNA。实验结果表明：抗原阳性鼠体螨10只组、5只组，抗原阴性鼠体螨10只组，游离螨50只组、10只组，抗原阳性鼠肺50mg、100mg组，所抽提RNA与探针杂交结果呈阳性，而抗原阴性鼠体螨5只组抽提RNA的探针检测结果呈阴性。
Dig-DNA probes of hemorrhagic fever with renal syndrome virus M S gene fragment were prepared by PCR and random primer labeling.The probes had speciality to HFRSV-RNA and could detect 10pg RNA by RNA-DNA dot-blot hybridization.The results of hybridization showed that positive reactions were found in 5, 10 chigger mites groups from antigenic positive rats,10 mites group from antigenic negative rats,10, 50 free chigger mites, and lung of antigenic positive mice 100mg,500mg.But negative reaction in 5 mites group from antigenic negative mice.
