β-淀粉样蛋白及D-半乳糖对大鼠学习记忆和胆碱酯酶活性的影响 被引量:25

摘要 目的 观察 β-淀粉样蛋白 (β- amyloid,Aβ)、D-半乳糖及两种药物联合使用时对大鼠学习记忆和大脑皮层胆碱酯酶活性的影响。方法 D-半乳糖腹腔注射〔5 0 mg/ (kg·d)〕,连续 6周。在腹腔注射的第 4周末 ,双侧海马内各注射 Aβ1μl(含量为 4μg)。手术后 2周 ,对各组大鼠进行开场行为、Y-型迷宫、被动回避测试及大脑皮层胆碱酯酶活性的测定。结果 与对照组相比 ,D-半乳糖组、Aβ组、D-半乳糖加 Aβ组大鼠的自主行为、学习记忆能力和大脑皮层胆碱酯酶的活性明显下降 ,尤其 D-半乳糖加 Aβ组明显。结论  D-半乳糖、Aβ对大鼠的行为及中枢胆碱能系统均有明显抑制作用 。 Objective To study the effect of β amyloid(Aβ),D galactose or Aβ plus D galactose on rat′s behavior and cholinesterase (CHE) activity in the cerebral cortex. Methods D galactose was daily injected into abdominal cavity for six weeks 〔50 mg/(kg·d)〕. Aβ was injected into both sides of hippocampus (each 4 μg/1 μl) at the end of the fourth week. Two weeks later, open field, Y maze and step through tests were evaluated, and CHE activity in the cerebral cortex was determinated. Results Rat′s spontaneous activities, learning and memory ability in D galactose group, Aβ group and D galactose plus Aβ group were worse than those in control group, and the D galactose plus Aβ group was the worst among the three groups; CHE activity of cerebral cortex was reduced in three exprimental groups as compared with control group, and that of D galactose plus Aβ group was significantly lower than that of D galactose group or Aβ group. Conclusion The combination of D galactose and Aβ was more potent on the behavior and central cholinergic system in rats.
