摘要
本研究通过分析茄子热胁迫抑制性消减杂交(SSH-cDNA)上调表达文库中获得EST序列以及基因组数据库中收集到的与耐热相关的EST序列,对茄子耐热相关的EST-SSR分子标记进行了搜索分析。在茄子热胁迫SSH-cDNA文库中找到了9个SSR位点,结合收集到的耐热EST序列中的25个位点,共设计了32对SSR引物。进一步的引物筛选结果表明,引物对EG6靶向文库EST序列YP249,在不同茄子材料中扩增显示多态性。YP249经实时荧光表达验证确定为热胁迫上调表达序列,考虑EG6的扩增位点为茄子热胁迫相关的SSR多态性候选位点。
The heat-stress related ESTs from the suppression subtractive hybridization cDNA(SSH-cDNA) library of heat-stressed eggplant and the genome database of NCBI website were analyzed and the heat-stress related EST-SSRs in eggplant were studied.9 SSR loci were found in the ESTs of eggplant heat-stressed SSH-cDNA library,the other 25 SSR loci were found in the genome database,a total of 32 pairs of SSR primers were designed.Further primer screening results indicated that the EG6 primers which targeted library EST YP249 showed polymorphism in different eggplant materials.The YP249 was then proved as an up-Regulated expression sequence in heat-stressed materials by real-time fluorescence,therefore,the EG6 amplification site was considered as a candidate SSR polymorphism locus which related to the thermotolerance of eggplant.
